This procedure allows you to isolate
chromosomal DNA by using ethanol. Thread like DNA
molecules can be seen with naked eye. The procedure
does not involve any steps of lysis or centrifugation.
Pre-isolated DNA is provided as a aqueous solution.
Visible DNA strands can be retrieved with ethanol
and a glass rod.
Time Needed to Complete the
Experiment:
Dissolving DNA in buffer: 10 minutes.
Addition of salt and precipitation of DNA with ethanol:
5 minutes.
Spooling DNA with glass rod: 5 minutes.
Run agarose gel electrophoresis (optional): 40 minutes.
Staining time:
2-3 hrs for methylene blue stain or instant results with
ethidium bromide stain.
