SPOOLING OF DNA (30 Preps)

Visualize thread like DNA by adding ethanol to DNA solution (enough for 30 students).
Standard DNA is also supplied for agarose gel.

Precipitate double stranded chromosomal DNA with      ethanol.
Visualize thread like DNA structures after precipitation.
Spool DNA threads with a sterile loop or glass rod.
Understand the physical nature of high molecular      weight chromosomal DNA.
Examine DNA under microscope.
Run agarose electrophoresis gel (optional) and stain      DNA.

This procedure allows you to isolate chromosomal DNA by using ethanol. Thread like DNA molecules can be seen with naked eye. The procedure does not involve any steps of lysis or centrifugation. Pre-isolated DNA is provided as a aqueous solution. Visible DNA strands can be retrieved with ethanol and a glass rod.




Time Needed to Complete the Experiment:
Dissolving DNA in buffer: 10 minutes.
Addition of salt and precipitation of DNA with ethanol:
    5 minutes.
Spooling DNA with glass rod: 5 minutes.
Run agarose gel electrophoresis (optional): 40     minutes.

Staining time:
2-3 hrs for methylene blue stain or instant results with ethidium bromide stain.

Kit includes following items:
Salmon Sperm DNA.
3M Sodium Acetate.
Glass rods.
Staining Solution (for optional agarose gel electrophoresis).
Standard DNA marker for gel electrophoresis.
Loading tips (for loading samples on a gel)
Blank sample for agarose gel as practice.
Gel running buffer.

Other Requirements:
Beakers, Ethanol, Water and Test Tubes.
