DNA mapping is a common technique used to determine the location of genes. Mapping of DNA by restriction enzymes not only provides the genetic information of DNA but it also provides the information about enzymatic cleavage sites which are used for subcloning DNA fragments into vectors. Subcloned DNA fragments are then used for DNA sequencing, protein expression and other molecular biology experiments. This experiment includes 8 different predigested DNA samples along with a standard DNA ladder. DNA fragments from each sample are separated on an agarose gel during electrophoresis. Depending upon the restriction enzymes used in each sample, a unique DNA pattern is obtained in a stained gel which is used to map the relative positions of restriction sites of the original DNA molecule. No human DNA is used in this experiment.

Time Needed to Complete the Experiment:
Casting the gel: 25 minutes.
Loading DNA samples: 10 minutes.
Running time for electrophoresis: 45 minutes.

Staining time:
2-3 hrs for methylene blue stain or instant results with ethidium bromide stain.

Predigested DNA samples (pre-cut with different restriction enzymes).
5 different samples (each 100 µl); Each sample contains different sizes of DNA      fragments.
Standard DNA (100 µl) for comparing fragments of various sizes.
Loading pipet of fix 20 µl volume. This pipet is extremely durable which can be used up to      5,000 times.
Microfuge tubes rack of 96 loading tips.
Heavy duty test tube rack containing 80 holes for microfuge tubes.
Agarose, enough for 10 gels
Concentrated electrophoresis buffer, enough for 10 gels
DNA staining solution.
Gel running buffer.
Measuring tubes (15 ml and 50 ml capacity) & microfuge tubes.
