Preparations of plasmid DNA are very
common procedures used in the field of DNA biotechnology.
This kit introduces the technique of isolation and
purification of plasmid DNA from transformed bacteria,
E. coli. Plasmid preps prepared with this method
are pure enough for cleavage with restriction enzymes.
Plasmid preps can also be used for transformation
experiments. Kit includes specially formulated proprietary
solutions which give maximum yield of plasmid preps.
Extraction of DNA does NOT require the use of phenol
or chloroform.
Time Needed to Complete the
Experiment:
Grow E.coli cells (16 hours) or use frozen cells.
Make pellet from grown culture: 5 minutes
Lysis of cells: 10 minutes.
Precipitation and purification of DNA: 20 minutes.
Run agarose gel electrophoresis (optional): 40 minutes.
Staining time:
2-3 hrs for methylene blue stain (instant results is stain
is used in the gel or staining with ethidium bromide stain).

E.coli cells (two sets of pre-transformed cells with plasmid
DNA).
Powder LB media for growth of E.coli cells (includes antibiotic
for E.Coli growth).
All reagents for plasmid preparations.
Standard DNA (100 µl) for comparing fragments of
various sizes.
Loading pipet of fix 20 µl volume. This pipet is
extremely durable which can be used up to 5,000
times.
Microfuge tubes rack of 96 loading tips.
Heavy duty test tube rack containing 80 holes for microfuge
tubes.
Agarose, enough for 10 gels.
Concentrated electrophoresis buffer, enough for 10 gels.
DNA staining solution.
Gel running buffer.
Measuring tubes (15 ml and 50 ml capacity) & microfuge
tubes.
Instructions sheet.
Blank sample for agarose gel as a practice.
