In this experiment different restriction enzymes are used to digest linear lambda DNA at unique nucleotide sequences. Bacteriophage lambda DNA has a linear structure with 48, 502 base pairs which gives a unique pattern of DNA fragments on agarose gel after cutting with restriction enzymes.. Different restriction enzymes recognize specific sequences of this large molecule of DNA. Following Agarose gel electrophoresis, staining of gel will yield distinct DNA bands depending upon the type of the enzyme used for cutting the DNA.


Time Needed to Complete the Experiment:
Digestion of DNA by restriction enzymes: about 1 hour. DNA can also be digested      overnight with enzymes. Cut DNA can be saved in freezer for future use.
Casting the gel: 25 minutes.
Loading DNA samples: 10 minutes.
Running time for electrophoresis: 45 minutes.


Staining time:
2-3 hrs for methylene blue stain or instant results with ethidium bromide stain.

Double autoclaved sterile water
Standard DNA for comparing DNA fragments.
Loading pipet of fix 20 µl volume. This pipet is extremely durable which can be used up to      5,000 times.
Microfuge tubes rack of 96 loading tips.
Heavy duty test tube rack containing 80 holes for microfuge tubes
Agarose, enough for 10 gels
Concentrated electrophoresis buffer, enough for 10 gels
DNA staining solution and gel buffer
Measuring tubes (15 ml and 50 ml capacity) & microfuge tubes.
