Plasmid DNA such as pUC vector is
very common vector which produces ß-galactosidase
after hydrolyzes of X-Gal. When a foreign DNA fragment
is subcloned into the pUC polylinker region, gene for
the ß-galactosidase interrupted, resulting in an
inability to hydrolyze X-Gal. In presence of X-Gal and
IPTG (inducer), such interruption of ß-galactosidase
gene results in the production of white colonies. In this
experiment both white and blue colony forming plasmids
are provided. Pre-transformed E.coli cells are provided
(no dry ice necessary for shipping) which are stable for
long time at room temperature. E.coli cells containing
plasmid are grown on bacterial plates containing Ampicillin.
The new traits of DNA are identifiable as blue or white
colony after the transformation process.
Time Needed to Complete
the Experiment:
Preparation of antibiotic resistant plates: About 40 minutes.
Plates containing agar media can
be stored in refrigerator for up to three months.
Inoculation of plates with pre-transformed plasmid DNA:
5 minutes.
Results:
After 16-24 hours of incubation of plates at 37 °C
or about two days of incubation at room temperature.
