Transformation leading to fluorescence red colonies: Fluorescent
protein are isolated from marine organisms, which are
widely used to study the localization for fusion proteins
in mammalian expression systems. In most of fluorescent
proteins, appropriate mutations are introduced to increase
the fluorescence intensity before cloning into appropriate
expression plasmids. After transformation, cells are selected
based upon the antibiotic resistance which is encoded
within the plasmid DNA. Surprisingly, these proteins do
not require any substrates, or cofactors to emit the light.
The DsRed™ Fluorescent Protein was isolated from
Discosoma- an IndoPacific relative of sea anemone and
coral. DsRed™ Fluorescent plasmid has been provided
to Science Vision International by Clontech - to be used
only for educational purpose.
Time Needed to Complete
the Experiment:
Grow E.coli cells (16 hours) or use frozen competent cells.
Preparation of small scale competent cells: 20 minutes.
Preparation of antibiotic resistant plates: About 40 minutes.
Plates containing agar media
can be stored in refrigerator for up to three months.
Transformation of plasmid DNA to frozen or fresh competent
cells: 30 minutes.
Inoculation of plates with transformed E.coli cells: 5
minutes.
Results:
After 16-24 hours of incubation of plates at 37 °C
or about two days of incubation at room temperature.
